So, I have a question. In the opening scene of Flash, the characters are discussing, very briefly, other superheroes.
Heroes mentioned, not necessarily by name, are Superman, Wonder Woman, Green Arrow, Canary & "Assorted Gotham People."
I wanted to have an element of interconnectivity from the start. My question is...
Is this legit? Can I do this? Or will it cock something up?

All of those are in continuity so it sounds fine to me. Power Girl and Great Machine would also be public superheroes, the latter of which is presumably retired unless Action Comics says otherwise.

OK. As I said, it's a really brief description of things anyway.
I mostly just want it to be a conversation to show peoples personalities before we go into the Origin of the Flash and the BIG OL" EXPLOSION.

My rule is if it was posted in a news paper or blog then a character who would keep up with such things would be fair game to use.

If all you're doing is referencing events that have already happened in continuity, then you don't need to run it past anybody.
If you want to do cameos or simply just run it past others for quality control, feel free to PM what you like to one or more admins and we'll be happy to look over it.

Excellent. I'm currently trying to work out if I should have a Flash already in existence or not...I've come unstuck a little bit, as I really want Teri to be the focus (although, Defo going to involve Jesse Quick now that she is free.)

I've seen it go both ways.
Writing an origin means you get to define a character, which can be helpful. And done well a very positive thing. However it can also be very slow and lead to readers complaining that "it's not X".
Writing an established character skips that part, and leads into sheer action. However you have to depend on fore knowledge. Depend on a history a reader might or might not know. It's a toss up.
The best thing I've seen, as a middle way, is a intro scene. More touching on the process. With Flash that might work.
